MnSOD activity and the effect of the Y34N mutation. (A) The increase in absorbance (absorbance units) at 420 nm of 120 mM Y34N MnSOD in the 2 ms after pulse radiolysis generated initial superoxide concentrations of 5 mM. (B) The same experiment at longer times. Solutions contained 30 mM formate, 50 µM EDTA, and 2 mM Taps at pH 8.2. The enzyme had been reduced prior to the experiment with 130 mM H2O2. The data are fit to eq 1eq 4 resulting in the entries to Table 2.
