Effect of divalent cations on the binding affinity of human Ago2 for various antisense oligonucleotides

The dissociation constants (Kd) for human Ago2 containing the various antisense oligonucleotide configurations in the presence of either Mg2+ or Mn2+ were determined as described under “Materials and Methods.” Sequences of the unmodified 19-nucleotide antisense RNA (19-as) and antisense RNAs containing 5′-hydroxyl (OH), abasic residue (p/a), or deoxyribonucleotides substitutions (dN). The errors reported for the Ago2 binding affinities are based on three trials.
