Inhibition of AuNP-HDAC8 by TSA. AuNP-HDAC8 conjugate was prepared with 3 μM HDAC8 and assayed for enzyme activity in the presence of varying concentrations of TSA. Activity was plotted as a function of TSA concentration. The fitted curve represents the best fit of the data for competitive inhibition model with Ki = 127 ± 18 nM. Ki for inhibition of free HDAC was similarly determined to be 131 ± 15 nM, in agreement with the reported value 19.
