DLC1 binding to phospholipids. (A) In 96-well plates, immobilized egg PC plus 15 mol% of the indicated lipids was incubated with hypotonic lysate from HEK293T cells transiently expressing GFP-tagged DLC1. Bound DLC1 was detected with GFP-specific antibody, followed by HRP-conjugated secondary antibody, and visualized with ABTS peroxidase substrate. Absorption was measured after 30 min at 405 nm. The mean of two independent experiments performed with triplicate samples is shown; error bars, SEM. (B) Immobilized PI(4,5)P₂ (15 mol% in PC) was incubated with hypotonic lysate from HEK293T cells transiently expressing GFP-tagged DLC1 wild-type (WT), ΔSAM, and ΔSTART proteins.
