The Stability of Designed TPRs. The thermal denaturation curves of the designed TPRs (MMY open circles, MLY closed triangles, and VMY open triangles) overlay well with that of TPR2A (open squares), indicating there is no destabilization due to introduction of the hydrophobic pocket. T-Mods MLY and VMY have an additional K5H mutation that had no effect on melting temperature. The constructs used for all of the other experiments discussed in this work do not have this mutation.
