Two Cys-rich domains are required to bind to immobilized LH. Man/GalNAc-4-SO₄-Fc (Mu1) and Man/GalNAc-4-SO₄-Fc (Mu11) were cotransfected in CHO/Tag30A cells and labeled with Trans[³⁵S]-label. Secreted Fc chimeras were isolated by protein A affinity chromatography. The Fc chimeras were incubated with Man-Sepharose in TBS containing 5 mM Ca²⁺ or LH-Sepharose in TBS containing 5 mM EDTA. Starting material for each matrix (Input), unbound (UB), and bound (B) fractions were examined by nonreducing SDS/PAGE. The positions of Man/GalNAc-4-SO₄-Fc (Mu1) 340,000 Da, Man/GalNAc-4-SO₄-Fc (Mu1/11) 220,000 Da, and Man/GalNAc-4-SO₄-Fc (Mu11) 98,000 Da are shown.
