MfdC shows gain of function in a translocase assay. A triplex-containing substrate consisting of a ³²P-labeled TFO bound to a supercoiled plasmid was incubated with 250 nM full length Mfd or MfdC, and reactions were initiated by the addition of 2 mM ATP. Samples were removed at certain intervals, quenched, and analyzed by gel electrophoresis. The data show the percentage of TFO displaced from the substrate at each time point, and are the average of three independent experiments, with standard deviations shown.
