A) ES cells cultured with or without retinoic acid for three days were subjected to real-time RT-PCR analysis. For differentiations, 1μM retinoic acid (RA) was used or LIF was withdrawn from the medium. ES cells were transfected with respective plasmids for three days before RA addition and selected with antibiotics for five days. Error bars represent standard deviation from three technical repeats. B) Embryoid body (EB) formation of control or Mbd3 knockdown stable cells. Stable cells (2×105) were suspension cultured in ES cell medium without LIF in 35 mm cell dishes.
