INS-1E cells were transfected with an empty vector (Control), with a plasmid encoding a shRNA directed against GFP (shGFP), RalA (shRalA) or PLD1 (shPLD1) [18]. Three days later the cells were incubated under basal conditions in a medium containing 2 mM glucose or were stimulated during 10 min with 20 mM glucose, 30 mM KCl, 10 µM Forskolin and 100 µM IBMX. At the end of the incubation period, cell lysates were prepared and assayed for PLD activity. Histograms show PLD activity (mU/50 µg of proteins) measured under basal and stimulatory conditions. The results are the means ± SD of three independent experiments. * p<0.05, one-way ANOVA.
