Function of leucocytes in wounded tissues. (a) Phagocytosis of damaged neutrophils by macrophages. Thioglycollate-induced peritoneal macrophages were collected from the peritoneal cavity of C57BL/6 mice as phagocytes. Zymosan-induced neutrophils were collected from the peritoneal cavity of Green mice. Neutrophils were heat-treated (100° for 10 seconds). They were opsonized (37° for 15 min) using serum from sham-operated or splenectomized mice. Then macrophages and neutrophils were mixed and analysed by fluorescence-activated cell sorting. Left panel, non-opsonized; middle panel, opsonized by the serum from sham-operated mice; right panel, opsonized by the serum from splenectomized mice. Representative results are shown from three independent experiments. (b) Expression of chemokines and cytokines in wounded tissues. RNAs were extracted from wounded tissues at each time-point. The expression of macrophage chemokines CCL2, CCL3, CXCL3 was analysed by reverse transcription–polymerase chain reaction. Results were shown as relative band intensity. Representative results are shown from three independent experiments.
