Enzyme activity of 2′5′AS in non-stimulated and IFN-α stimulated beta TC3 and alpha TC3 cells. Beta TC3 (a) and alpha TC3 (b) cells were incubated in the presence or absence of 500 U/ml of IFN-α. After the indicated time periods, cell lysates were prepared. The lysates were incubated with 0.15 μCi α ³²P-ATP for 120 minutes at 30°C in the presence of 100 μg/mL poly(I:C), and then separated by ascending TLC. The generated 2′5′A were quantitated by PhosphorImager analysis. Enzyme activity was expressed in units per milligram (n = 4, standard deviations varied between 6–13%).
