CTL responses to the HIV-1 Gag448-457 peptide in HLA-A2 transgenic mice in the presence of different amounts of gp96 as an adjuvant. Four groups of mice were immunized with 50 μg of the 10-mer peptide and different amounts (0, 10, 20, and 50 μg) of gp96. Splenocytes were obtained 7 days after the last immunization. (A) ELISpot assay. Splenocytes at 106 cells per well were tested in the presence of 10 μM of the peptide. Bars represent the means from three independent experiments. *, P < 0.05. (B) Tetramer staining. Splenocytes isolated from the immunized mice were stained with HLA-A2/kb2-microglobulin-10-mer peptide tetramer along with anti-CD8-FITC and anti-CD3-PE-Cy5 antibodies. The analyzed splenocytes were gated on CD3+, CD8+, and tetramer-positive cells. Numbers are percentages of tetramer-positive cells within the CD8+ and CD3+ T lymphocytes.
