Sre1 pathway mutants have defects in Sre1 production. Log-phase cells of B-3501A and the indicated mutant strains were incubated with 21% (+) or 1% (−) oxygen for 4 h at 30°C in YES medium. Then, the cultures were shifted to 21% oxygen for 15, 30, or 60 min in YES medium. Whole-cell extracts were prepared and subjected to immunoblot analysis using anti-Sre1 antiserum. P and N denote the Sre1 precursor and its cleaved nuclear forms, respectively.
