Ty3 particles associate with GLFG domains of Nups in vitro. GST fusions to Nups or domains of functional FG repeats were expressed in E. coli and purified using glutathione-Sepharose beads, and similar amounts were used as bait in the affinity capture assay with extracts of yTM443 cells induced to express Ty3 for 24 h (see Materials and Methods). Elution fractions were separated using sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and immunoblot analysis was performed using anti-GST (top panel), anti-CA (middle panel), and anti-IN (bottom panel) IgG.
