Carboxyfluorescein (CF) efflux induced by mastoparan (masL) from LUVs of POPS/POPC 50:50 by direct addition of masL (0.5 μM) to vesicles with encapsulated dye, with lipid concentrations of 30, 50, and 100 μM (fastest to slowest). The gray lines are experimental data (averages of 2–5 traces in 2 independent samples) and the thin solid lines are the fits of the graded kinetic model to the data. The fits use the same kon as for masX, a koff 30 times larger than for masX (Table 2), and the insertion and efflux parameters listed in Table 3. The experiments were performed at room temperature (about 22°C).
