Lower bounds on the number of unique TCRβ CDR3 sequences in the CD8+CD45RO+/ and CD4+CD45RO+/ T cell compartments in two young, healthy, CMV-seronegative adult males were calculated using an analytic solution to the ″unseen species problem″ (see “Methods”). GA1 and GA2 indicate whether the sequence data were obtained before (GA1) or after (GA2) the implementation of technical enhancements to the Illumina Genome Analyzer that increased the maximum sequence read length from ∼ 36 to ∼ 54 nucleotides.
