Relative yields of wild-type and mutant human and guinea pig 11β-HSD1 proteins in the bacterial expression system. Proteins were produced and purified as per Materials and Methods. Levels of purified protein produced are shown in mg protein per liter LB broth and are representative of more than one experiment. Introduction of glutamate at position 275, 278, and 279 in the guinea pig sequences, and 278 in the human sequence, dramatically increased the yield of recombinant 11β-HSD1 protein.
