Survivin-specific siRNA (Sri-2) inhibits survivin expression determined by western blots. Control: transfected with transfection reagent only; scraSRi: transfected with scramble RNA; and Sri-2: transfected with Sri-2. B. MCF-7 cells were plated in 12-well plates (105 cells/well) and grown for 24 hours. Cells were sequentially transfected with or without survivin siRNA (SRi-2) or scramble siRNA (scraSRi) for 24 hours and then treated with or without T138067 (100 nM) for 24 hours as shown, followed by photographing. C. Experimental conditions were the same as in A, cell death was analyzed by trypan blue exclusion assays as described in the Methods, data were plotted as a histogram and each bar represents the mean ± SD from three independent assays.
