Frequency of Suc^r isolates from plasmid cointegrates

Complementing plasmid pSW2.6 containing PA906 and parent plasmid pBBR1MCS were mated into cointegrates SW129 and SW323 that contain plasmid pSW906KO integrated into the chromosome. Resultant Cb^r Gm^r, Km^r, and Cm^r isolates plus and minus complementation were grown overnight at 37°C in LB broth containing Gm and Cm without Km. Dilutions were plated onto LB agar containing Gm and Cm with and without 5% (vol/vol) sucrose. Frequency of sucrose resistance is calculated as the ratio of the number of isolates on agar containing 5% (vol/vol) sucrose to the number of isolates on agar without sucrose.
