Effects of mrp4 knockdown on the levels of Cox-2 and PGE2 in wild-type MEF cells. A, siRNA-mediated knockdown of mrp4 in mrp4+/+ MEF cells. Mrp4+/+ MEF cells were transfected with the nontarget sequence shRNA (control), mrp4-shRNA 1, or mrp4-shRNA 2 and then grown for 72 h. Total protein was isolated from cells and analyzed for mrp4, Cox-2, and actin protein by Western blotting, with relative quantitative levels of mrp4 and Cox-2 given below the blots. B and C, Levels of extracellular and intracellular PGE2 in mrp4-knockdown MEF cells. After 48 h of transfection as described in A, the growth medium was removed and replaced with fresh complete medium and cells were grown for another 24 h. The medium and purified cell lysates were assayed for the levels of extracellular and intracellular PGE2, respectively; the levels of PGE2 are expressed as picograms per milligram of cellular protein.
