THP-1 cells (2×10⁶) were pretreated with inhibitors of (A) p38 MAPK (SB202190; 0.1 µM), (B) ERK1/2 (PD98059; 30 µM), (C) JNK (Inhibitor II; 10 µM), and (D) Akt (LY294002; 10 µM) for 30 min and then stimulated with LPS (1 µg/ml) in the presence or absence of CO gas (250 ppm) for 2 h. DMSO (0.1%) vehicle was used as control. IL-1β production was measured in supernatants by ELISA. Results are mean ± SEM of three to five independent experiments.
