VLC depletion potentiates the induction of a peptide specific immune response. (A) MAFIA mice were challenged with 5 x 10⁶ ID8-C3. After 6 weeks, mice were injected with AP20187 (AP20187) or injection solution (Vehicle) for every other day for 5 days. On the fifth day, mice were injected with 2 µg of SIINFEKL peptide/10 µg of lipopolysaccharide/100 µg of agonistic αCD40 antibody. As a control, naiveMAFIAmice were also immunized at this point. (B) After 72 hours, AP20187-treated (AP20187), injection solution-treated (Vehicle), and immunized naive mice were killed and spleens were stimulated with PMA/ionomycin. After 5 hours, cells were harvested and stained for CD8 and intracellular IFN-γ. Cells were analyzed by FACS, and the percentage CD8⁺IFN-γ⁺ cells compared with naive immunized mice were determined. (C) Mice were treated as in panel B, except stimulated with 1 µg/ml SIINFEKL peptide and analyzed as a percentage of the CD8⁺IFN-γ⁺ cells produced from naive mice.
