ACSL3 knockdown decreases lipogenic gene expression. Rat primary hepatocytes were plated at 0.5 × 106 cells/22-mm well and transfected with 1 μg of siRNA per well. After 50 h of siRNA transfection, total RNA was isolated and mRNA expression of lipogenic genes (A) or transcription factors and co-activators (B) were quantified using quantitative RT-PCR and normalized to RPL-32. C, proteins were harvested at 50 h after transfection and subjected to Western blotting for measurement of ChREBP.
