CDK1 is required for nocodazole-induced Rad53 phosphorylation. A, exponentially growing cdc28-as1 cells were treated for 3 h (lane N) with nocodazole (15 μg/ml). 1NMPP1 (dissolved in dimethyl sulfoxide (DMSO)) was then added to one-half of the culture. Samples were taken at the indicated time points to test FACS profiles and Rad53 phosphorylation by Western blot with Mab.EL7 monoclonal antibodies. B, exponentially growing cell cultures of wild type and isogenic clb2Δ, clb5Δ, and clb2Δclb5Δ strains were treated for 3 h (N) with nocodazole (15 μg/ml).
