Model showing the concentration of PKD, Gαq, PLCβ, and PKCδ at the NHERF-1 scaffold. Stimulation of GPCRs leads to activation of PLCβ, resulting in the local generation of DAG. DAG within the plasma membrane results in the recruitment of both PKD and its upstream kinase PKCδ by binding to their C1 domains. PKCδ is in the vicinity of NHERF-1 by associating with actin, which is linked to NHERF-1 via ezrin. PLCβ and Gαq are known to be associated with NHERF-1 via a PDZ interaction. The enrichment of PKD and these upstream components at NHERF-1 results in a concentrated PKD response downstream of activating stimuli.
