CD8°OT-1 transgenic thymic lobes were cultured in the presence of media or 50 μM E1, T1, or OVA peptides. Thymocytes from cultured lobes were harvested on day 7, counted, and analyzed for expression of CD4, CD8, and Vα2 by flow cytometry. (a) CD8 versus CD4 expression for each culture condition.
