FACS gating strategy used to purify mouse wild-type LT-HSCs for transplantation. Mouse LT-HSCs were purified with the phenotype cKit+Thy1.1(lo)Lin(-/lo)Sca1+Flk2- (KTLS cells) (red box). Multipotent progenitors (MPPs) are also highlighted as the cKit+Thy1.1(lo)Lin(-/lo)Sca1+Flk2+ population (black box in the far right KLS plot). E12.5, E15.5, and E18.5 fetal liver (FL) samples and 8 week bone marrow (BM) are shown. The percentage of total white blood cells (WBCs) within the gates is shown, with the exception of the KLS plots (far right) which show percentage of the KLS population. PI, propidium iodide.
