GS-HNE stimulation of human PMNs: generation of superoxide anion and modulation of CD11b. Human PMNs were isolated from healthy volunteers as described in Methods and incubated with indicated concentrations of GS-HNE (A) for 10 min at 37°C. The superoxide dismutase (SOD)-inhibited reduction of ferricytochrome c was monitored spectrophotometrically at 550 nm. The increase in superoxide generation stimulated by GS-HNE (10 µmol·L−1) was inhibited by SOD (B). fMLP (50 nmol·L−1) was used as a positive control. (C) represents the modulation of CD11b expression upon treatment with GS-HNE (10 µmol·L−1; 15 min) as determined by FACS. Results are presented as mean ± SEM (cells from three separate donors), *P < 0.05 (vs. GS-HNE by unpaired Student's t-test) and **P < 0.01 (vs. saline), ***P < 0.001 (vs. saline) by one-way anova.
