A 3-aa change in the NES of mouse MTF-1 (mMTF-1) to the corresponding human residues confers high metal inducibility. MTF-1 null cells (dko7) were transfected with a reporter plasmid under the control of four tandem MREs, a reference plasmid, and the indicated wild-type or mutated MTF-1 construct of mouse or human. Thirty-six hours after transfection cells were treated with 100 μM zinc sulfate for 4 h. RNA was isolated from the cells and quantified by the S1 nuclease protection assay.
