Overexpression of GRP78 does not affect DENV production. (A) 293 cells were transiently transfected with pEFIRES-P-BiP to express GRP78 or pEGFP. At 48 h posttransfection, the cells were lysed and the lysates were subjected to Western blotting with rabbit anti-GRP78 and detection of complexes with goat anti-rabbit IgG-HRP conjugate and chemiluminescence. The Western blots were reprobed with mouse anti-actin and visualized with goat anti-mouse IgG-HRP and chemiluminescence. (B) At 48 h posttransfection, the cells were infected with DENV at an MOI of 1 or mock infected. At 24 h p.i., the supernatant was sampled and assayed for infectious virus by plaque assay. The results represent means plus standard errors of the mean from three independent transfection/infection experiments.
