Reaction of BFDC with PAA monitored by stopped-flow PDA. (A) Difference spectra showing PHB–ThDP formation in the reaction between BFDC and PAA. BFDC (33.2 μM active-site concentration) in buffer B was mixed with an equal volume of 20 mM PAA in the same buffer at 30 °C. The reaction was monitored in the indicated wavelength range for 32 s, and spectra were recorded every 5 ms. (B) Rate of formation of PHB–ThDP. Reaction conditions as in A. Data were fitted to a single exponential: A = A0(1 − exp(− kt)) with SigmaPlot vs 7.01. The rate constant k−5 refers to that in Scheme 1.
