Fig. 6 — Effect of various pharmacological agents and hydrogen peroxide on PMCA. (A) Neurons were exposed to DQ (50 μM), BSO (100 μM) or PQ (50 μM) for 24 h or pretreated for 1 hour with MnTMPyP or MnTBAP (50 μM) prior to exposure to PQ. The cells were lysed in 3 mM Tris-HCl, pH 8.0, and 10 micrograms of protein separated and probed with pan PMCA antibody (1:1000). (B) Neurons were exposed to H₂O₂ (50 μM) for 18h, proteins were separated under reducing vs non-reducing conditions, and the membranes probed with the pan PMCA antibody (1:1000).
