Agarose gel electrophoresis of total RNA extracted from N. benthamiana inoculated with CMV-Y + Y-satRNA, T1C2(C2-T2)C3 + Y-satRNA, T1T2C3 + Y-satRNA, or V-TAV(J) + Y-satRNA. Total RNAs were extracted from the upper leaves for detection of Y-satRNAs as described by Masuta et al. (12). The positions of the single- and double-stranded Y-satRNAs are indicated. A discrete band of single-stranded Y-satRNA was observed in coinoculations with T1C2(C2-T2)C3, but the double-stranded Y-satRNA (dsRNA) was difficult to visualize in the same preparations. This phenomenon is similar to the observation of Moriones et al. (13), who found that in contrast to CMV, V-TAV does not induce accumulation of large amounts of double-stranded RNA.
