Both the FID and SID regions of Opy2p are required for normal response to hyperosmolarity. (A) Yeast cells (opy2Δ ssk1Δ) transformed with OPY2 plasmids with indicated deletions were assayed for the ability to grow on the hyperosmolarity media indicated. (B) Dose–response curves of Opy2p alleles. Yeast cells as in A) bearing the Opy2p alleles indicated were assayed for transcriptional reporter (8xCRE-CYC1-LacZ) activity under indicated osmotic stress conditions for 1 h before the β-galactosidase activity was measured. (CT)2 indicates an Opy2p construct having a duplicated cytoplasmic tail (see text for details).
