In vitro evaluation of 2-NBDG cellular uptake. (a) Fluorescence microscopy of 2-NBDG uptake in HT-29 cells after incubation in 1.0-mM 2-NBDG for 30 min. (b) Cellular uptake of 2-NBDG by HT-29 cells is concentration dependent, with uptake increasing monotonically as the concentration of 2-NBDG in the culture media increases. (c) Correlation of FDG and 2-NBDG uptake inhibition by excess glucose. Transport of both 2-NBDG and FDG is reduced into MCF-7 cells by excess glucose in the growth media, with both molecules showing similar decrements in intracellular uptake as the extracellular glucose concentration is increased.
