Immunoblots of M. maripaludis ΔflaK cells complemented with flaK or pibD. (A) Immunoblot of M. maripaludis crude membrane fractions from wild-type, ΔflaK, and ΔflaK(pHW40::flaK) cells using anti-FlaB2 antibodies (1/2,500) as primary antibodies. Cells were grown in nitrogen-free medium supplemented with alanine (promoter on) or NH4Cl (promoter off), as indicated. (B) Immunoblot of M. maripaludis crude membrane fractions from wild-type, ΔflaK, and ΔflaK(pHW40::pibK) cells using anti-FlaB2 antibodies as primary antibodies. Cells were grown in nitrogen-free medium supplemented with alanine (promoter on) or NH4Cl (promoter off) and at 30°C or 37°C, as indicated. Wild-type and ΔflaK cells were grown at 30°C, but identical blots were obtained when these cells were grown at 37°C. U, unprocessed flagellins; P, processed flagellins.
