Table 3.

Percent of FtsZ-Bound Nucleotide That Is GDP, GTPase Activity, and Polymer Measured by Light Scattering Measured at Different GTP Concentrationsa

GDP percentage GTPase activity activity (GTP min−1FtsZ−1) light scattering intensity
40 μM GTP 28 4.3 35
60 μM GTP 35 5.4 43
80 μM GTP 47 6.1 50
120 μM GTP 50 6.4 49

The FtsZ was the unlabeled wild type, at a concentration of 20 μM. The GTP concentrations in the first column include 20 μM nucleotide that accompanied the 20 μM FtsZ. The GTPase activities reported in the second column were measured at 30 °C, whereas values in Table 1 were measured at 25 °C. For light scattering measurements, the excitation and emission of the fluorometer were both at 350 nm.
