Model of 3BP2-mediated NFAT activation after BCR stimulation. BCR stimulation induces the sequential activation of non-receptor type of PTKs Lyn, Syk, and tyrosine phosphorylation of Tyr183 in 3BP2. Phosphorylated Tyr183 directly binds to the SH2 domains of Vav1 and PLC-γ2. 3BP2-SH2 domain directly binds to BLNK. In addition, 3BP2-SH2 domain was reported to associate with Syk, Vav1, and PLC-γ2. 3BP2-mediated signaling complex facilitates to the activation of NFAT.
