FVIII·vWF complex inhibits RANKL-induced osteoclastogenesis. A, RAW 264.7 cells were cultured for 5 days in the presence or not of 100 ng/ml human RANKL (hRANKL), 100 ng/ml OPG, and 1 or 2 units/ml FVIII·vWF complex. After May Grünwald/Giemsa staining. B, purified human CD14+ monocytes were cultured for 15 days in the presence of 25 ng/ml human macrophage colony-stimulating factor and 100 ng/ml human RANKL and 1 unit/ml FVIII·vWF. TRAP coloration was performed at the end of the culture period. C, purified human CD14+ monocytes were cultured for 15 days in the presence of 25 ng/ml human macrophage colony-stimulating factor and 100 ng/ml human RANKL with or without 50 ng/ml OPG and 1 unit/ml FVIII·vWF. Multinucleated TRAP-positive cells were counted under a light microscope. A and C, results are expressed as the number of multinucleated cells (more than three nuclei) per well; each value represents the mean ± S.D.
