The PVL histidine (H)-to-arginine (R) mutation does not affect pore formation in human PMNs. LukS-PV^R, LukS-PV^H, LukF-PV, LukD, and Hlg-B were produced as recombinant proteins. The indicated amounts of LukS-PV^R and LukS-PV^H were combined with each F component (LukF-PV, LukD, and Hlg-B) and immediately used in the leukotoxicity assay as described in Fig. 1. The results obtained with the same blood donor, are expressed as the mean of three or four experiments ± the standard error of the mean. In the analysis of variance test, P values were not significant between LukS-PV^R and LukS-PV^H alone or in combination with LukF-PV, LukD, or Hlg-B.
