PAC-1 Fab binding to CHO cells expressing αIIbβ3 and αIIbβ3(D723R). PAC-1 Fab binding levels were determined for cells expressing wild type αIIbβ3 (solid lines) or αIIbβ3(D723R) (dashed lines) in the absence (solid squares) or presence of Mn²⁺(open circles). In this and subsequent figures, binding is expressed as a ratio of specific PAC-1 immunofluorescence (PAC-1 MFI) over total integrin detected by the αIIbβ3 antibody HIP8 (HIP8 MFI). Specific PAC-1 binding is the total PAC-1 immunofluorescence minus that seen in the presence of EDTA. Values shown are the mean ± S.E. (error bars) from 3 independent experiments.
