Schematic diagram of Cx43-eYFP indicating predicted topology of Cx43 and the position of eYFP and its fusion by an eight amino acid linker (SRDPPVAT) to the C terminus. Cx43 is predicted to span the plasma membrane four times and has cytoplasmically-located N and C termini. The locations of the amino acid mutations examined in this study are indicated. F52dup is located in the extracellular domain, L90V is located in the second transmembrane domain, and Y286A and LSYTRF are located in the cytoplasmic tail.
