Celecoxib attenuates the inhibition of ovCOX-1 by aspirin. (A) ovCOX-1 (2.0 μM) was preincubated with the indicated concentrations of aspirin and celecoxib at 37° for 1 h, and a small aliquot assayed for COX activity using an O2 electrode with 40 μM AA as the substrate. When present, the concentrations of aspirin and celecoxib in the O2 electrode assay chamber were 7.5 μM and 0.38 μM, respectively. Values are represented as mean ± SEM, n = 3, and ∗∗P < 0.0001 vs. control using one-way ANOVA. (B) ovCOX-1 (2.0 μM) was not treated or was preincubated for 2 h at 37° with the indicated inhibitors (0.5 mM aspirin, 25 μM celecoxib, and 25 μM ibuprofen). Aliquots of the enzyme samples were assayed for COX activity as indicated above. Values are the average of triplicate determinations ± SE.
