IL-3 deprivation causes a decrease in Bcl-2 mRNA levels in both WT and p53^−/− FDM cell lines. (A) Three independent WT and p53^−/− FDM cell clones were deprived of IL-3 for 6 or 18 hours. RNA was extracted, and gene profiles were analyzed with the use of the 6-chip Illumina expression array. Heatmap depicts the expression of key Bcl-2 family genes. (B) FDM cells of the indicated genotypes were deprived of IL-3 over a 48-hour time course. Lysates were prepared, and proteins were resolved on SDS-PAGE. Western blots were probed with antibodies to Bcl-2, Bcl-x_L, Mcl-1, Bak, Bax, Bid, Bim, and β-actin (loading control).
