Cysteines of TMD 4 and 10 are required for full charge movement. A, NLC curves constructed from mean fit parameters show that C192S/C196S and C415S prestin have reduced area under the curve, indicating reduced charge movement compared with WT prestin. V½ and z for these mutants are consistent with WT measures (see Figs. 3 and 5).
