Matriptase·HAI-1 complexes are shed into extracellular milieu following acid-induced activation. 184 A1N4 cells were preincubated with basal media (activation −) or pH 6.0 buffer to induce matriptase activation (activation +). After 20 min the cells were washed and then incubated in basal media for the indicated periods after which cell lysates and conditioned media were collected and analyzed by immunoblotting for total matriptase and activated matriptase, as indicated.
