Stable gene expression in human MDM. (A) Primary cultures of human MDM were transduced with the defective lentiviral vector (MOI = 50) at day 7 in vitro, and GFP expression was then examined 46 days later (one of four representative experiments is shown). (B). RT-PCR detection of transgene expression in DLV-transduced human MDM. Total RNA was isolated from both DLV-transduced and normal MDM and RT-PCR amplification was conducted with the use of GFP-specific primers flanking a 373-bp fragment. The PCR products were subjected to 2% agarose gel electrophoresis.
