Activation of NF-κB by TLR2 mutants in response to a synthetic tri-acylated lipopeptide presented under a complexed form to sCD14. 293 cells were incubated with increasing concentrations of Pam3CSK4-sCD14 complexes for 5 h at 37 °C. At the end of the incubation, the cells were lysed and assayed for expression of a NF-κB reporter construct as described under “Experimental Procedures.” The results are expressed as the means ± S.D. and are representative of two independent experiments. On each panel, responses of 293 cells expressing the indicated mutant (○) is compared with that of cells expressing wild type TLR2 (●).
