mTOR selectively regulates transcription factor activity during OL differentiation. Total RNA was extracted from OPCs after 3 d of differentiation ± 15 nm rapamycin. A, Olig2, Sox10, or Nkx2.2 mRNA levels were quantified using validated QuantiTect primer assays (Qiagen). Rapamycin treatment did not significantly affect alter the mRNA levels of Olig2, Sox10, or Nkx 2.2. B, Id2, Id4, or Tcf4 mRNA levels were quantified using validated QuantiTect primer assays (Qiagen). Inhibition of mTOR caused a 2.7-fold, 3.8-fold, and 4.6-fold increase in the mRNA levels of Id, Id4, and Tcf4, respectively (Id2, *p = 0.027; Id4, *p = 0.0259; Tcf4, *p = 0.0238).
