Measurements were done in duplicate. The first two bars are negative controls, where either DCoH or HNF1 had been left out. Additions to the incubation step are as follows: 0, no addition; DTT, 1 mM DTT; BH4, 100 μM BH₄; BH4+DTT, 100 μM BH₄/1 mM DTT (DTT was added to keep BH₄ reduced); BH2, 25 μM 7,8-BH₂.
